Abstract

Genetic variations in the FOXO1A gene, single nucleotide polymorphisms, have increased the risk of type 2 diabetes mellitus (T2DM). Considering the earlier reported SNPs in FOX 01 gene showing association with type 2 diabetes, the aim of this study is to evaluate similar correlation. The population focused in this study is of North Indian region as earlier reported work mainly emphasized on South Indian region population, The study performed on 100 case-control population of T2DM groups for checking the association of SNP in the FOXO1A gene with T2DM and other risk factors of its complications. All the subjects were genotyped for the FOXO1A, SNP by polymorphism chain reaction (PCR) and restriction fragment length polymorphism (RFLP) methods, using hnf1 restriction enzyme with three fragments (133 bp, 85 bp and 48 bp) of the PCR amplicon to differentiate between homozygotes and heterozygotes to define the genotypic and allelic frequency. The result of our study suggest no significant association between SNP1–(rs2297627) and SNP2 -(rs2721068) and the risk of type 2 diabetes mellitus. In other words, foxo1a gene did not show a major diabetes gene pool capacity in north Indian population, the alleles were protective. The odd ratio of both SNPs signifies that there is no correlation between genotype and disease.
